It depends upon your setup. If you use one for OS and other for backup and temp operations it will be fast. And also if you are using IDE interface HDD then sometimes it increase your speed than normal or gets slowdown like I told it depends upon setup.
If both are same size and brand, you can setup in RAID 0 mode if mobo supports, it will be faster than normal operations especially in copying and moving files, Applications responding etc,...
If it is older system check your mobo specf's to see any Size limitations for HDD. And also make sure you have minimum of 512 Mb or More for better response. More size more seek time in some occasions.
