As a newbie to DNS, could anyone give me a bit of help.
For example: Name Server 1: Run01.mydomain.com Name Server 2: Run02.mydomain.com I think I have to create an ‘A’ record, and reverse lookup zone. I have a small confusion here, can I use my name server name(Run01) as my computer name.

Here's the basic layout of a zone:
http://serialcoders.sytes.net/BIND/basiczone.com.zone And you can have your computer name be whatever you want. It has no effect on DNS if you're using BIND or djbdns or tinydns.
