Folder permissions?

When working with a file in my editor I want to be able to view it in the browser. If the file is in my root web directory (d:\ActiveWebFiles\) I can see it in the browser with no problem. If, however, it's in a subdirectory (d:\ActiveWebFiles\CDEssentials\AdminNew\index.php), the browser doesn't "find" the file and I get a "file not found" error.

Could someone please give me a two second tutorial on permissions and setting them in IIS?
