I don't think you should ever assume a user is using 1024x768. Some older people have large monitors, but still use 800x600 due to their poor eyesight. If you use a fixed width site, make sure the design doesn't break if the user increases the size of the font. More importantly, let the user change the size of the font the typical weight. Use em or percentages for font sizes if at all possible.
